Keith & Co., page 108 mentions “Apocynin” root having long been used by native Americans prior to the arrival of Europeans, so its use as a medicinal herb in North America dates back many centuries. As of 1883, it was becoming studied more closely as an impure substance extracted from the roots of certain small plants that grow in Northern US and Canada. About 8 years later, a chemist named Otto figured out how to synthesize Apocynin in the lab, but scientists of that time had their doubts about whether the laboratory version was truly the same as the plant-derived version. In 1908, an English chemist (Finnemore) decided to find the answer. He obtained 40 Kg of the plant roots and after great time & effort, he extracted 80 grams of pure Apocynin from it. (40,000 grams yielded only 80g of the Apocynin,,,a very low 0.2% yield.) Reference: Journal, Chemical Society of London, Vol.93, part 2, pp.1513-19. The Englishman then performed exhaustive tests that proved the laboratory-made Apocynin is identical in every physical, medical and biochemical way to the Apocynin that occurs naturally. MEDICINAL PROPERTIES OF APOCYNIN TAKEN ORALLY Let's start with a little background about our immune systems. White blood cells circulating in our blood are our 1st line of defense against infection. There are several types of them, --most prominent are the neutrophils and the macrophages. When white blood cells encounter a 'bad' invasive cell (such as a fungus, bacteria or virus) they perform some chemical tricks involving oxygen, enlisting the oxygen to help them attack the invasive foreign cell. In a way, Oxygen has a split personality. An individual oxygen atom does not like to be alone. It is chemically ‘aggressive’. It wants to join/react with things around it. Fortunately, when two individual oxygen atoms join together, they stabilize each other by forming a molecule of the life-sustaining oxygen that we all breathe. So, a single oxygen atom is ‘hyper’, but two of them joined together into a molecule are ‘calmer’. Our immune system takes advantage of this “split personality” of oxygen, and employs several methods to revert it's calm version into its aggressive, reactive “hyper” version, to fight infection. The advance guard of our immune systems are the specialized, infection-fighting white blood cells called macrophages. Each patrolling macrophage contains an enzyme called “NADPH oxidase”. If a macrophage senses a foreign invader (such as a harmful virus, bacteria or fungus), the NADPH oxidase within it will instantly trigger the production of SUPEROXIDE, --a specialized molecule that is armed with the ‘aggressive’ version of oxygen that wants to react with something. You could visualize this immune response as being akin to a patrolman who wakes up and releases a specialized type of ‘attack dog’. (We’ll discuss more about superoxide, below.) It is important to understand that, in attacking the invader, that 'attack dog' (a macrophage or some other type of white blood cell) is also creating a certain level of inflammation. The larger the number of white blood cells that become present to attack an invader, the larger the amount of inflammation that occurs. If an entire army of white blood cells get called into action, this large response can unintentionally result in levels of inflammation that are harmful. This is where Apocynin can step in to help keep things more balanced. When invading pathogen cells (fungus, bacteria, virus) become spotted and attacked by the "advance guard" macrophages, the macrophages release small protein molecules called cytokines, which signal the more-common variety of white blood cells (neutrophils) to join the attack. If the pathogen cells are rapidly multiplying, our various white blood cells release even more amounts of cytokines, signaling even more of their fellow white blood cells to join the fray. Apocynin is a very remarkable, and very multi-faceted, medicinal herb substance. Since Apocynin is great at keeping the “wild side” of oxygen well under control, it's also great at controlling another variety of ‘wild’ oxygen (called “free radicals”). Free radicals can be formed when incoming solar radiation (UV rays) whack into our cells the wrong way. And our cells (especially the mitochondria in their nuclei) can sometimes accidentally produce them on their own, in the course of going about their routine business. In the USA, the FDA considers it to be a dietary / herbal supplement (not a drug) for which optimum human oral dosage levels have not yet been established. Thus, there is no “pharmaceutical” form of Apocynin recognized by the United States. (A foreign pharmaceutical manufacturer may describe it as such, but they are describing it in terms of the standards in their own country. Since the other country's standards can vary significantly from US FDA standards, they are not applicable here.) Although optimum human oral dosage levels have not been established, various studies involving laboratory animals have indicated that relatively low oral dosage levels of Apocynin are sufficient to produce effective results. When researchers are conducting tests to determine optimum oral dosage levels, they will often start with a lower daily dose. For example, they may start with a 50 mg dose, then gradually increase the daily dosage in small increments to determine a level where the best results are being achieved --without overdoing it. However, if taking an oral dose of Apocynin, it may be better to take it earlier in the day, instead of taking it near bedtime. ------------------------------------------------------------------------------------------------------------ ABOUT THE TOPICAL (SKIN) APPLICATION OF APOCYNIN On April 3, 2019 a group of Japanese researchers (Tokyo) announced their discovery that Apocynin can rejuvenate skin stem cells and maintain their continued activity. The 26-page scientific publication, with the title,,, 'Stem cell competition orchestrates skin homeostasis and ageing', was published in Nature. For a good 2-page summary of the Nature publication, do a web search for: "Anti-aging discovery could lead to restorative skin treatments", published in Scientific American, on April 4, 2019 (written by Dana Smith). In their April 3, 2019 publication, the researchers describe experiments in which they divided mice into three groups and then “wounded” them by surgically removing small, identical-sized areas of skin from their tails, --after having first established that the skin from a mouse tail is closely similar to human skin. They then applied solutions to the tail wounds in each group. (Let’s call them Groups A, B & C.) Group A’s wounds basically received a placebo solution (called a “vehicle” solution). Group B’s wounds received a solution containing a small amount of a synthetic lab-created substance called “Y-27632”. Group C’s wounds received a solution containing the nature-based Apocynin. As expected, the “vehicle” solution did very little. But they found that the Y-27632 solution and the Apocynin solution were each separately able to stimulate production of a vital skin protein called COLLAGEN 17. In Groups B and C, the wounds healed much better and faster, with much less scarring. In some ways, Y-27632 was superior to the Apocynin, but it is a rare research chemical that is not available to the public, and it is extremely expensive. Apocynin, a natural substance, came in a “close second”. It is an available, low-cost alternative to the lab-created Y-27632. They discovered that Apocynin increases your own body's production of COLLAGEN 17, which is the essential nutrient for the stems cells that create fresh skin cells. Thus, they had discovered a way to actually rejuvenate and maintain the deep, fundamental cellular activity & structure of the skin. Apocynin is functional,,, not a cosmetic! It can serve to create more ‘food’ for the skin stem cells and therefore help restore/maintain the actual foundation of the skin. To confirm the validity of the results observed in the tests they conducted with the skin of mice, they performed numerous ‘parallel’ tests with human skin cells grown in glass lab dishes. This produced very similar results, thus establishing that Apocynin (and Y-27632) can stimulate / restore increased stem cell activity in human skin. In the skin, the aging process causes a gradual breakdown of an intricate choreography that begins with a horizontal bottom ('basal') layer of stems cells that are always competing with each other, in a process that 'weeds out' the weaker stem cells. The healthiest stem cells are the ones that are getting the most nourishment by being firmly anchored to the bottom basal layer (i.e. the microscopic “basement floor” of our skin), where they keep producing fresh new skin cells. Those new cells then s-l-o-w-l-y develop and migrate up through various layers of our epidermis, until they reach our skin surface as adult skin cells to replace the ones that are always "flaking off" of us. Over time, such things as UVB sun radiation or the gradual accumulation of ‘free radical’ damage within our cells causes a depletion of Collagen 17, and this causes some skin stem cells to gradually begin 'losing their footing', instead of remaining firmly anchored to the ‘basement floor’ layer. This causes them to become less active, and they stop producing fresh skin cells. They can hang on in this weakened dormant state for quite a while, --still present, but also not producing new skin cells. At some point, those stem cells finally get completely detached. Once this happens, they are permanently lost. This slow and steady attrition of stem cells means our skin is producing less replacement skin cells. With the skin cell production line becoming progressively ‘undermanned’, our skin gradually develops the tell-tale signs of aging. The researchers discovered that Apocynin reinforces the health of the stem cells that were already still doing well. But more importantly, they found it can also restore many of those weaker 'borderline' stem cells that had been doing little more than taking up room,,, “sleeping on the production line”, so to speak. With those stem cells now ‘re-awakened’ and actively producing fresh skin cells again, the entire skin structure is fundamentally improved. Apocynin helps support the maintenance and the rejuvenation of stem cells in skin tissue. The following hypothetical example is intended to help explain some further details. But, if you are the sort of person who prefers to read all the exacting technical details, please read the full 26-page paper, referenced above. It is truly seminal, a very well-conceived and well-executed piece of research. Older persons will naturally have a greater population of weak, inactive stem cells than younger persons do. As a hypothetical illustration, let's say 15% of a 70-year-old person’s original skin stem cell population has been gradually, permanently lost over the years, never to return. And maybe another 15% of that person's skin stem cells have become that weak, inactive population that is still present, but not producing. At this point, the aging process may have effectively reduced his/her ability to produce fresh skin cells by 30% (15% gone forever + 15% inactive). Only 70% of the original stem cells are still producing fresh skin cells. Far too few fresh cells are now being produced to replace old, dying cells,,, and the effect becomes seen in the form of skin that is becoming thinner & more fragile, resulting in sagging skin, wrinkles, eye bags, age spots, slowed wound healing, more scarring, etc. In our example, the 70-year-old taking Apocynin for the first time can never replace the 15% of stem cells that had already been permanently lost forever. He/she got started too late to save those 15%. But thanks to the Japanese research group's discovery, we now know that Apocynin is able to restore Collagen 17 to levels where it can provide essential nourishment to the 70% of skin stem cells that were already healthy, and can keep them healthy. The Apocynin appears also able to re-awaken many of that other 15% of stem cells that had become non-productive, and can help keep them from lapsing back into inactivity again. They join the other 70% that had already still been productive, so now you have more stem cells making more fresh skin cells. By restoring the vital Collagen 17 levels, the Apocynin can make a significant difference and can help 'hold the line' against further skin aging. In this sense, Apocynin is therefore truly ANTI-AGING. Apocynin can reasonably be expected to need time to work. It can take several months for fresh 'newborn' skin cells to slowly migrate from the parent skin stem cells, journey up thru the epidermis layers, and finally make their arrival at the skin surface. While the Japanese skin research is certainly tantalizing, the extent of Apocynin's potential to actually make human skin appear "younger" is unknown at this point in time. It would be very unrealistic to expect Apocynin to suddenly reverse years of skin aging overnight to suddenly make you look years younger. However, the research seems to suggest that Apocynin does have the potential to slow the gradual appearance of further skin aging. This can be significant for anyone, but perhaps more significant the younger you are. If you are currently say age 45 and looking into the mirror some future day 25 years from now, you may be very glad that you started applying the inexpensive Apocynin skin solution way back in year 2024. You just may look appreciably younger than your fellow 70-yr-olds who are around you in year 2049. Hair follicles are another type of skin tissue, so it’s no surprise that some scientists (the stereotypical bald men with glasses?) have looked into its potential to restore hair. (Even older scientists can dream, right?) Unfortunately, the results were not encouraging, -at least not encouraging for older, long-bald individuals whose hair follicle stem cells became permanently lost long before this Apocynin discovery was ever made. You cannot restore a follicle stem cell that is no longer even present. But, this may not be the case for younger persons in whose scalps the dormant hair follicle stem cells may have just started to slow hair production (producing early signs of hair loss). Although dormant, those follicle stem cells are still physically present, so the research seems to indicate that at least some of them could be potentially recoverable. Regular topical applications of the aqueous Apocynin solution to the scalp could help support renewed activity of the dormant follicle stem cells. This could possibly slow further hair loss. Again, the Apocynin research was published fairly recently, and the aging process is very slow & gradual; there hasn’t yet been enough time to gauge what Apocynin can - or can not - do in this regard. It is worth noting that the same Japanese research group (and others) continue to investigate whether Apocynin can have beneficial effects on any other types stem cells found within our numerous organs and body systems. Given its safe low toxicity, its proven beneficial properties, and its relatively low cost,,, Apocynin would appear to be a sensible addition to a personal health regimen. MAKING THE TOPICAL SOLUTION : The April 3, 2019 publication says the Japanese researchers applied a 100 micromolar solution of Apocynin in phosphate-buffered saline solution (PBS) with an 0.5% wt/wt amount of DMSO added. (SEE OUR PHOTO of the formula, as published). In plainer terms, this is roughly equivalent to dissolving about 1 mg of Apocynin into two ounces of hot distilled water and then adding just one single drop of DMSO, and just 1mg of non-iodized table salt . At this low concentration, 10 grams ($20) of Apocynin would be sufficient to produce 156 gallons of the topical solution, at the strength they used. (1 gram = 1000 milligrams). (10 gram = 10,000 milligrams). This works out to only about 13 cents a gallon. At this low concentration, the distilled water would actually cost you more than the little speck of Apocynin that you are dissolving into it. NOTE : The single drop of pure DMSO per two ounces of solution is added to slightly increase skin permeability and allow the Apocynin to penetrate the skin just deeply enough to reach the layer of skin stem cells, and stay there. A larger amount of DMSO would just uselessly carry the Apocynin entirely thru your skin, and into your bloodstream. (In our experience, the German “Heiltropfin” brand of 99.5% DMSO is the most reliable. It is sold in small, brown glass bottles at an affordable price, and has a nice glass “pipette bulb” type of eyedropper built into the bottle’s screw lid.) In the Japanese research results, the beneficial skin effect is shown as being “dose dependent” (i.e. the higher the concentration, the greater the benefit). However, they did not describe having reached the point where they felt they had achieved an 'optimum solution concentration'. This leaves open the possibility that a slightly higher concentration could perhaps produce greater results. If a ‘stronger’ solution were to be desired, it would simply be a matter of dissolving a greater amount of Apocynin into the hot ‘water’. As an example, 10 mg could be dissolved into the 2 ounces, instead of just 1 mg. (Along with the same single drop of pure DMSO.) This would make a solution that is 10x’s more concentrated than used by the Japanese experimenters. If used at this higher concentration, 10 grams of Apocynin would be enough to make 15.6 gallons of the skin solution, and each single gram (a couple dollars’ worth) could produce 1.56 gallons of it (= 200 oz). The skin solution still remains very inexpensive. It is definitely worth looking into. Among our PHOTOs, you will see one that shows 100mg of Apocynin in the tiny measuring spoon, filled slightly less than level. (If the tiny spoon is filled level smooth, it will contain about 130 mg.) You will also notice another photo showing just 10mg in the same tiny spoon. As you can see, 10mg is a very small amount. If a person were to decide to make a solution that is 5x’s more concentrated than the Japanese group employed (instead of 10x’s), he/she could dissolve the 10mg Apocynin into 4 oz. of the hot distilled water, instead of 2 oz. water, thus diluting it. (Since you would now have 4 oz. of solution, you would now be using two drops of the pure DMSO, and 2mg of the salt.) NOTE #1: Although hot distilled water is used for preparing the skin solution, it must be allowed to cool to room temperature before being applied to the skin. NOTE #2 : If a person were considering applying a stronger Apocynin solution to his/her skin, it would be prudent to start by first applying it to some “test area” of the body that is less visible.
